The Dr Reddy's Institute of Life Sciences was conceived in the context of globalization, to foster research competence in India and to bridge the divide between academic research and the pharmaceutical & biotechnology industries.  The institute was incorporated as a non-profit entity under Section 25 of the Indian companies act, facilitating operational autonomy. Construction of the DRILS building comprising 44 labs and an administrative block and spanning ~130,000 sq.ft. in an area, began in 2004 and was completed in January 2007.

PROJECT TITLE
A mechanistic study of the recruitment of the nuclear RNAi machinery for de novo establishment of heterochromatin domains in S.pombe

Qualification
- Post Graduate Degree in Basic Science with NET qualification (M.Sc. Biochemistry/Biotechnology/Genetics/Molecular Biology etc)
- Graduate Degree in Professional Course with NET qualification (B.Tech. Biotechnology or similar)
- Post Graduate Degree in Professional Course (M.Tech. Biotechnology or similar)
NETs accepted: CSIR-NET including LS; GATE, JAM, GPAT, BET, JEST, JGEEBILS, ICMR-JRF and ICAR- AICE Candidates with a keen interest in basic science research and prior hands-on experience with molecular Biology, biochemistry or microbiology techniques are preferred. A possibility of PhD enrollment exists, however it is not guaranteed

PROJECT SUMMARY
In eukaryotes, heterochromatin domains play a key role in packaging, protecting as well as regulating the expression of the genetic material. In S.pombe, the nuclear RNA interference (RNAi) machinery is involved in the formation of heterochromatin and it appears likely that a similar process may function in mammalian cells. In this proposal, we aim to address the following questions: How does the first wave of the RNAi machinery localize to the repeats, what role do the DNA or RNA sequences play in this process? What is the kinetics of this process and is there an order of assembly and a time scale associated with it? This information is important especially in the context of data that implicates the nuclear RNAi pathway in the setting up facultative heterochromatin in mammalian cells, and playing a role in development, chromosome segregation and surveillance. The successful outcome of this study will identify sequence elements that are essential for de novo establishment of heterochromatin and silencing. In addition, we will also be able to generate a kinetic roadmap of how the RNAi machinery is recruited and silencing established.
